II. Indications

  1. HIV Infection (part of combination therapy)
    1. High risk of induced HIV resistance
    2. Never use NNRTIs as monotherapy in HIV Infection

III. Mechanism

  1. See Non-Nucleoside Reverse Transcriptase Inhibitor (NNRTI)
  2. Second generation NNRTI

IV. Dosing

  1. HIV Infection in Adult (or child weight >35 kg), in combination with other Antiretrovirals
    1. Take 100 mg orally daily

V. Adverse Effects

  1. See Non-Nucleoside Reverse Transcriptase Inhibitor

VI. Safety

  1. Avoid in Lactation
  2. Unknown safety in pregnancy (registry exists)

VII. Drug Interactions

  1. Rifabutin
    1. Increase Doravirine frequency to 100 mg orally twice daily
  2. CYP3A4 Inducers
    1. Avoid strong CYP3A4 Inducers with 4 weeks of Doravirine
    2. Examples include Carbamazepine, Phenobarbital, Phenytoin, Rifampin
  3. NNRTIs
    1. Do not coadminister with other NNRTIs including Efavirenz, Etravirine, Nevirapine
