III. Mechanism

  1. Placental Alpha Microglobulin-1 Rapid Immunoassay (PAMG-1 Protein Marker Assay)
  2. PAMG-1 is 1000 to 10,000 times more concetrated in amniotic fluid than in cervical or vaginal secretions

IV. Technique

  1. No vaginal exam should be done prior to testing
  2. Sterile Dacron swab is placed at the posterior vaginal fornix for 1 minute
  3. Swab is placed in solvent vial for 1 minute
  4. Test strip is placed in solvent and result is available in 5 minutes

V. Efficacy

  1. Test Sensitivity: 95.7%
  2. Test Specificity: 100%
  3. Contrast with Test Sensitivity of 78.1% for combined standard methods
    1. Vaginal Fluid Ferning
    2. Vaginal Fluid pH (Nitrazine)
    3. Vaginal Pooling
