- Severe Familial Hypercholesterolemia (e.g. LDL Cholesterol >190 mg/dl)
- Enlicitide is the first oral PCSK9 Inhibitor as of 2026 (others in class are Parenteral)

IV. Medications
- Enlicitide (Lipfendra) tablets
- Store at room Temperature in original container with desiccant (tablets are moisture sensitive)
- Cost/year: $3800 at time of release in 2026 (50% of injectable PCSK9 Inhibitors)
V. Dosing
- Enlicitide (Lipfendra) 20 mg orally daily
- Take with water or plain Coffee or tea (no additives)
- Delay other food or beverage until at least 30 minutes after taking Enlicitide
- When transitioning from injectable PCSK9 Inhibitor
- First dose of Enlicitide should be on the day when the scheduled injectable would have been due
VI. Efficacy
- LDL Cholesterol lowered 60% when added to a Statin (at 20 mg dose, similar to injectable PCSK9 Inhibitors)
- Cardiovascular outcome studies not available at time of release in 2026
