III. Mechanism

  1. See Neurokinin-1 Receptor Antagonist
  2. Blocks the neurokinin-1 receptor (a tachykinin)
  3. Crosses the blood brain barrier to inhibit CNS Substance-P activity, reducing Motion-Induced Vomiting

IV. Dosing

  1. Tradipitant 85-170 mg orally 60 minutes before triggering event (e.g. boating)
  2. Take on an empty Stomach
  3. Do not repeat dosing for 24 hours

V. Efficacy

  1. Decreases Vomiting (NNT 4)
    1. However, minimal effect on motion-induced Nausea

VI. Adverse Effects

  1. Minimal sedation (contrast with Antihistamines such as Meclizine)
  2. Expensive (in 2026, each 85 capsule costs $255)

VII. Drug Interactions

  1. Strong CYP3A4 Inhibitors (e.g. Clarithromycin)
    1. Increases Tradipitant adverse effects
